Coiled-coil domain-containing protein 7 (CCDC7) is a protein-coding gene frequently implicated in cancer biology[1][2][3]. Its linear form is upregulated in cancer stem cells, such as CD133-positive colorectal cancer, and elevated in advanced stage and high-grade cervical cancers, where it promotes cell proliferation and engages the JAK-STAT3 signaling pathway through upregulation of IL-6 and VEGF[2]. CCDC7’s circular RNA (circCCDC7) encodes a tumor suppressor peptide (circCCDC7-180aa) shown to inhibit prostate cancer progression by suppressing cell migration and invasion, and correlates with improved prognosis when highly expressed[1]. The mechanisms of CCDC7 therefore vary by RNA and protein form and by tumor context, with the linear RNA/protein acting as an oncogene in multiple cancers and the circular RNA product functioning as a tumor suppressor in prostate cancer. No drug interactions or direct inhibitors are currently known in the literature, and CCDC7/circCCDC7 is considered a promising prognostic biomarker and potential therapeutic target, especially in oncology.[1][2][3]
