Overview

Coiled-coil domain-containing protein 74A is a human protein encoded by the CCDC74A gene, located on chromosome 2q21.1[1][3]. The protein is 420 amino acids in its longest isoform and contains two coiled-coil domains of unknown function; it is predicted to localize mainly to the nucleus but may shuttle to the cytoplasm[1]. CCDC74A is most highly expressed in the testis and trachea, with moderate expression in several other tissues including lung, brain, prostate, spinal cord, bone marrow, ovary, thymus, and thyroid gland[1]. It is believed to play a role in developmental and differentiation pathways, particularly during spermatogenesis, and may regulate gene expression[1][3]. Interactions have been documented with lysine demethylase KDM1A and SH2D1A, suggesting involvement in chromatin modification and immune pathways[1]. There are hints of clinical relevance: CCDC74A promotes cell proliferation in androgen-dependent prostate cancer models and its expression is influenced by dihydrotestosterone exposure[1]. The protein is evolutionarily conserved with one major paralog (CCDC74B) but has limited orthology outside primates[1][3][7]. Genetic or protein function annotation remains largely descriptive, with no direct evidence for targeted therapeutics or established safety concerns at present[1][3][7].
