Overview

Coiled-coil domain-containing protein 77 (CCDC77) is a long coiled-coil protein predominantly encoded in the human genome and expressed in the nucleus and/or cytoplasm of all tested tissues[6][3][9]. CCDC77 plays a role as a structural component of the A–C linker, an essential element for centriole stability. The protein localizes specifically at the distal appendages of mature centrioles and is involved in the integrity and duplication of centrioles—key processes in cell division and centrosome organization. Functional studies indicate CCDC77 interacts with microtubules and co-localizes with other centrosomal proteins such as WDR67 (TBC1D31) and MIIP, possibly forming a microtubule-associated complex fundamental for the cohesion and anchoring of microtubule triplets during centriole formation[2][7][9]. There is no current evidence identifying CCDC77 as a direct therapeutic target, such as a receptor, enzyme, transporter, or signaling molecule; instead, its main function lies in fundamental cellular architecture and microtubule-organizing center biology. No known small-molecule drugs or biologics target CCDC77, and it is not recognized as a clinical biomarker or a source of specific toxicity or safety concern[3][9][6]. In summary, CCDC77 is a coiled-coil domain-containing, centrosomal, and structural protein crucial for centriole integrity, with no direct roles as a drug target or clinical biomarker established in the literature.
