Overview

Coiled-coil domain-containing protein 80 (CCDC80) is a secreted, extracellular matrix-associated protein encoded by the CCDC80 gene on human chromosome 3[3][4][5]. It is involved in cell adhesion, matrix assembly, and the regulation of extracellular matrix organization[4]. CCDC80 has heparin and fibronectin binding activities and is predicted to be present in the extracellular region and basement membrane, functioning upstream of responses to bacterial infection[4]. CCDC80 acts as a tumor suppressor in several cancers: its expression is frequently downregulated in colorectal, thyroid, pancreatic, melanoma, and gastric cancers, while its ectopic expression inhibits cancer cell growth and migration, likely through upregulation of E-cadherin and modulation of apoptosis[2][5]. In mouse models, deficiency of CCDC80 disrupts cholesterol and bile acid metabolism, promoting hypercholesterolemia by impairing the hepatic alternative bile acid synthesis pathway[1]. CCDC80 is not currently considered a direct drug target, nor are there known drugs with established mechanisms of action acting through CCDC80. However, it is regarded as a candidate biomarker for tumor suppression and metabolic disorders and may aid in patient selection or monitoring for some cancer subtypes[2][5].
