Cold-sensitive ion channels are a subset of transient receptor potential (TRP) family ion channels primarily responsible for the molecular detection of cold in sensory neurons. The key members—TRPM8 and TRPA1—are nonselective cation channels activated by cool and cold temperatures, as well as chemical agents (menthol for TRPM8, pungent irritants for TRPA1). Channel activation results in cation influx, membrane depolarization, and neuronal signaling to the central nervous system, where cold is perceived. These channels are implicated in both physiological (thermosensation) and pathological (pain, inflammation) processes. While “cold-sensitive ion channels” as a collective target is physiologically meaningful, structured information should resolve to the individual channel subtype whenever possible
Agonists (e.g., menthol) induce or potentiate channel opening, mimicking cold or enhancing cold sensation. Antagonists block activation and reduce cold-induced or pain signals.
