Collagen alpha-6(VI) chain is a large extracellular matrix glycoprotein encoded by the COL6A6 gene, featuring multiple von Willebrand factor type A domains and a characteristic triple-helical region. It belongs to the expanded collagen VI family, which comprises six genetically distinct alpha chains (α1 to α6), and participates in the structural organization of connective tissues such as skin, muscle, cartilage, bone, and cornea. COL6A6 may display tissue-restricted expression, with particular abundance in regions of the skin and neuromuscular junction. While the fundamental role of collagen VI is mechanical stabilization of the extracellular matrix and cell anchorage, recent studies suggest possible involvement of COL6A6 in skin homeostasis and pathology (e.g., atopic dermatitis). However, COL6A6 is not currently classified as a receptor, enzyme, or classic therapeutic target. Its molecular assembly and structure resemble those of collagen alpha-3(VI) chain, suggesting possible evolutionary and functional parallels.
