Collagen bioactive peptides are short peptide fragments (2-45 amino acids) derived from enzymatic hydrolysis of collagen proteins, rich in hydroxyproline, proline, and glycine, which allow them to resist digestion and enter the bloodstream intact via transporters like PepT1. They stimulate synthesis of extracellular matrix proteins in tissues such as skin, cartilage, joints, tendons, ligaments, bone, and skeletal muscle by activating pathways like mTOR, promoting cell differentiation (e.g., fibroblasts, myoblasts, osteoblasts), and upregulating gene expression for tissue remodeling. Specific di- and tripeptides like Pro-Hyp and Hyp-Gly exert systemic effects including antioxidant, antihypertensive, and wound-healing activities, while supporting hyaluronic acid production and satellite cell activity for muscle hypertrophy. In disease contexts, they show potential benefits for osteoarthritis, osteoporosis, skin aging, and bone loss by enhancing collagen production and inhibiting proteoglycan degradation, though evidence for many uses remains preliminary. Unlike traditional therapeutic targets such as receptors or enzymes, these peptides function primarily as nutritional supplements rather than direct drug-binding sites, with absorption leading to elevated plasma levels of marker peptides like Pro-Hyp peaking 60-120 minutes post-ingestion.
