The **collagen type I alpha-2 chain** is a polypeptide encoded by the COL1A2 gene. It is one of the two major chains (along with the alpha-1 chain) that assemble into the triple helix forming type I collagen, the most abundant structural protein in the human body. Type I collagen is fundamental to the integrity and strength of connective tissues such as bone, skin, tendon, and the sclera of the eye. The alpha-2 chain combines with two alpha-1 chains to form a procollagen triple helix that is processed and crosslinked into fibrils, which give tissues their tensile strength. Mutations in COL1A2 are associated with heritable connective tissue disorders, notably various forms of osteogenesis imperfecta and Ehlers-Danlos syndrome. While critical for tissue structure and health, the alpha-2 chain is not a typical drug target but rather a structural molecule whose defects underlie several genetic diseases.
