Collagen type XXIV alpha 1 chain (COL24A1) is a minor fibrillar collagen protein mainly expressed in the developing and mineralizing skeleton, particularly in osteoblasts during bone formation. It helps regulate type I collagen fibrillogenesis, thus playing a significant role in extracellular matrix organization and bone mineralization. COL24A1 has binding motifs for integrins and mediates its effects through interactions with integrin β3 and modulation of the TGF-β/Smad signaling pathway. Its expression promotes osteoblastic differentiation and mineral matrix deposition, and silencing COL24A1 impairs these processes in vitro. Disease associations remain largely uncharacterized, though preliminary links include Anthracosilicosis and osteochondrosis. There is little evidence at this time that COL24A1 is a direct therapeutic target or that there are drugs acting on it.
