Colon motility refers to the coordinated contractions and relaxations of the smooth muscle layers in the colon that propel fecal material toward expulsion. This process involves complex interactions between intrinsic enteric neural circuits, extrinsic autonomic nerves (pelvic and splanchnic), local hormones such as cholecystokinin, circadian clock genes like period 1/2 (*per1*, *per2*), nitric oxide signaling via neuronal nitric oxide synthase (nNOS), immune mediators, and gut microbiota composition. Distinct regions along the proximal-distal axis exhibit unique structural and neurochemical properties influencing region-specific motor patterns. Disruption in these regulatory mechanisms can result in clinical disorders such as constipation or diarrhea seen in IBS or IBD patients. However, "colon motility" itself is not a discrete therapeutic target but rather an emergent property regulated by multiple molecular targets within neural-hormonal-microbial networks.
