Colon osmotic activity refers to the osmotic gradient that drives water movement across the colonic epithelium. This gradient is primarily established by active absorption of electrolytes—especially sodium through epithelial sodium channels (ENaC)—and the selective permeability of colonic epithelial cells. The process involves a coordinated system of channels and transporters, such as CFTR, ENaC, NKCC, and NHE, and is also influenced by solute concentration generated by organic acid metabolites from gut microbiota. Drugs such as osmotic laxatives work by increasing the osmolality of colonic contents to promote water retention in the stool and relieve constipation. Dysregulation of colon osmotic activity contributes to diseases such as diarrhea, constipation, IBS, ulcerative colitis, and cystic fibrosis. Note: "Colon osmotic activity" is not a well-defined pharmacological target such as a receptor or enzyme, and should not be listed as a canonical molecular entity. For drug targeting or biomarker purposes, specify the individual molecular components (like CFTR, ENaC, etc.) that regulate colonic osmotic gradients.
Increase stool water content by raising colonic lumen osmolality, thereby drawing water in through osmosis. Retarding absorption of water by maintaining high solute concentrations (osmotic laxatives resist absorption, trapping water in stool).
