Colony stimulating factor 3 receptor (CSF3R), also known as granulocyte colony-stimulating factor receptor (G-CSF-R) or CD114, is a cell-surface transmembrane protein that serves as the receptor for granulocyte colony-stimulating factor (G-CSF/CSF3). This cytokine-receptor interaction is essential for the regulation of production, differentiation, and function of neutrophilic granulocytes in the bone marrow. Mutations in CSF3R are implicated in several hematologic disorders, including chronic neutrophilic leukemia. Recombinant G-CSFs like filgrastim act through this receptor for therapeutic stimulation of white blood cell counts. Targeted therapies against aberrant CSF3R signaling—such as JAK inhibitors—are under investigation for malignancies driven by activating mutations.
