The Colorectal carcinoma-associated cell surface antigen recognized by mAb 1A3 is a lipid-based molecule, likely a glycolipid or neutral lipid, that is highly enriched on the surface of human colorectal adenocarcinoma cells [1, 6]. It was identified using the murine monoclonal antibody 1A3, developed at Washington University School of Medicine, which shows high specificity for colorectal tumors compared to normal tissues [1, 4]. The antigen is characterized by its solubility in the methanol phase of a Folch extract and its presence in most primary colorectal tumors and many metastases [6, 22]. This target has been extensively studied for diagnostic imaging and therapeutic applications, particularly in radioimmunoscintigraphy (RIS) and radioimmunoPET using isotopes such as Copper-64 and Indium-111 [4, 9, 29]. Clinical trials have demonstrated its utility in detecting small occult tumor foci in the abdomen and pelvis, although challenges such as the development of human anti-mouse antibodies (HAMA) and non-specific uptake in the liver have been noted [9, 33].
