LAVA-051 is a bispecific gamma-delta T-cell engager (Gammabody) that simultaneously binds to the Vγ9Vδ2 T-cell receptor and CD1d, a protein involved in presenting lipid antigens that is frequently expressed on the surface of various hematological tumor cells (LAVA Therapeutics, 2024). Interleukin-2 (IL-2) is a critical cytokine that promotes the proliferation, differentiation, and functional activation of T cells and natural killer cells (National Cancer Institute, 2024). The combined use of LAVA-051 and IL-2 is a therapeutic strategy designed to exploit the synergy between targeted T-cell engagement and systemic immune stimulation. In this regimen, LAVA-051 directs Vγ9Vδ2 T cells to attack CD1d-positive malignant cells, while IL-2 provides the necessary signals to expand the effector T-cell population and sustain their cytotoxic activity within the tumor microenvironment (de Bruin et al., 2021). This combination aims to improve clinical responses in patients with B-cell chronic lymphocytic leukemia, multiple myeloma, and acute myeloid leukemia by overcoming T-cell exhaustion and enhancing the durability of the anti-tumor immune response (LAVA Therapeutics, 2024). Preclinical data suggest that IL-2 significantly augments the degranulation and cytokine production of T cells engaged by LAVA-051, potentially leading to more robust clinical outcomes (de Bruin et al., 2021).
LAVA-051 acts as a bispecific engager bridging Vγ9Vδ2 T cells to CD1d-expressing tumor cells to induce lysis, while Interleukin-2 acts as a growth factor to enhance the expansion and effector functions of the activated T cells.
