COMMD10 is a scaffold protein within the Commander complex, composed of ten COMMD family members and other structural partners, which plays a critical role in intracellular transport, cell cycle regulation, immune response, and copper metabolism. It has demonstrated tumor suppressive activities in cancer, especially by inhibiting the NF-κB signaling pathway and enhancing apoptosis, and is implicated in increasing radiosensitivity in hepatocellular carcinoma by disrupting copper and iron balance. COMMD10 also regulates the immune response by controlling inflammasome activity and inflammation in monocytes and macrophages. Its network of interactions and co-expression with genes related to RNA processing and m6A methylation suggest additional roles in RNA biology and gene regulation. Although direct drug interactions are not established, COMMD10 is recognized as a potential diagnostic and therapeutic target in oncology and immunology.
COMMD10 functions by inhibiting NF-κB pathway activation (blocking p65 nuclear translocation and IκB degradation) when overexpressed or upregulated. It enhances radiosensitivity by inhibiting HIF-1α and disrupting copper-iron balance in tumor cells. Additionally, it stimulates apoptosis/cell death pathways, particularly Bcl-2/Bax/caspase signaling, and inhibits cell proliferation and invasion, demonstrating tumor suppressive activity.
