COMM domain-containing protein 2 (COMMD2) is a member of the COMMD family of proteins characterized by a conserved C-terminal COMM domain and an N-terminal α-helical domain, enabling it to form dimers and multiprotein complexes such as the CCC and Commander complexes that are essential for endosomal recycling and membrane protein trafficking[1][5]. Predominantly cytoplasmic, COMMD2 is implicated in regulating the surface expression of certain membrane receptors and may also influence sodium transport by interacting with epithelial sodium channel complexes[1][5]. Elevated expression of COMMD2 has been observed in multiple cancers, including hepatocellular carcinoma and bladder cancer, where it supports cell proliferation, migration, tumor progression, and is associated with immune cell infiltration and worse prognosis[2][3][4][6]. Recent studies suggest COMMD2 is a promising prognostic biomarker and potential therapeutic target in cancer, while its precise interacting drugs and detailed mechanisms of action remain to be elucidated[2][4].
