Overview

COMM domain-containing protein 3 (COMMD3) is a member of a conserved protein family (COMMD1–10) characterized by a C-terminal COMM domain enabling participation in hetero-oligomeric complexes such as the Commander complex[1][3][6]. COMMD3 acts as a central adapter in endosomal recycling, functioning both within the Commander protein trafficking complex (where it is essential for stability and activity of the CCC [COMMD/CCDC22/CCDC93] subcomplex) and independently by binding and stabilizing the small GTPase ARF1 via its N-terminal domain[1][2][3][4]. Independent of Commander, COMMD3 regulates specific trafficking routes for membrane proteins, such as the transferrin receptor (TfR), and is differentially required compared to other COMMD family members[1][2][3]. COMMD3 is also implicated in α-granule biogenesis in platelets and may influence copper homeostasis and transcriptional regulation[4][5][6]. Loss or altered expression of COMMD3 has been linked to increased tumor aggressiveness in some cancer models, particularly in breast cancer[5][6]. There are currently no known drugs that directly target COMMD3, and its role as a direct therapeutic target has not been established.
