Overview

COMM domain-containing protein 5 (COMMD5) is a member of the COMMD protein family and a scaffold component of the Commander complex, which is essential for endosomal recycling of transmembrane cargos and receptors. It plays a crucial role in maintaining epithelial cell phenotype, regulating cell proliferation, migration, and differentiation, as well as modulating the ErbB signaling pathway in some cancer contexts. COMMD5 expression is implicated in the progression or suppression of several cancers, including clear cell renal cell carcinoma, prostate, ovarian, breast, and lung cancers, with both under-expression and amplification linked to poor prognosis or tumor development. Initial identification of COMMD5 as HCaRG (hypertension-related calcium-regulated gene protein) highlights roles in kidney function and cardiovascular regulation. The protein’s position on chromosome 8q24.3 may influence its expression and function in disease, and COMMD5 expression and mutation analysis has potential diagnostic and prognostic value, especially in renal malignancies. Note: COMMD5 is not a typical drug target (such as a receptor, ion channel, or enzyme), but is involved in regulatory complexes that influence disease-related cellular pathways. No drugs are currently reported to directly target COMMD5.
