COMM domain-containing protein 6 (COMMD6) is a small, highly conserved, ubiquitously expressed protein that primarily consists of a COMM domain, characteristic of the COMMD protein family. It functions as an endogenous inhibitor of NF-κB signaling, modulating the ubiquitination and degradation of NF-κB subunits and thereby regulating inflammatory and cell survival pathways. Mutations in key residues of its COMM domain can abolish this inhibitory effect, leading to increased NF-κB activity. COMMD6 is also involved in endosomal receptor sorting (through participation in the CCC-WASH axis), in RNA processing and ribonucleoprotein biogenesis, and in regulation of copper homeostasis as part of the CCC complex. Its expression and modulation of these pathways have been linked to tumorigenesis and possibly other pathologies, though there are currently no drugs directly targeting COMMD6 and it is not established as a therapeutic target.
