COMM domain-containing protein 7 (COMMD7) is a member of the COMMD family, characterized by a conserved COMM (copper metabolism gene MURR1 domain) domain at the C-terminus and a helical N-terminal domain[4]. COMMD7 participates in multimolecular complexes such as the Commander (CCC) complex, which plays roles in endosomal trafficking, protein recycling, and cellular homeostasis[3][4]. COMMD7 specifically regulates signaling pathways involved in cell proliferation and cancer biology, including acting as a modulator of the NF-κB pathway—either promoting or terminating NF-κB activity depending on cellular context and interacting partners[1][2]. Overexpression of COMMD7 is associated with adverse prognosis in several cancers, such as hepatocellular carcinoma and pancreatic ductal adenocarcinoma, and may serve as a biomarker in acute myeloid leukemia[1][2]. Emerging research suggests COMMD7 acts as a multifunctional regulator at the interface of inflammation, cancer signaling, intracellular membrane traffic, and immune response[5][1][2]. No known drugs or small molecules directly targeting COMMD7 have been reported.
