COMM domain-containing protein 8 (COMMD8)

COMM domain-containing protein 8 (COMMD8) is a member of the conserved COMMD protein family, characterized by a COMM domain at the C-terminus. COMMD8 acts as a scaffold protein within the commander complex, which is essential for the endosomal recycling of transmembrane cargo proteins. The commander complex includes the CCC complex components (COMMD, CCDC22, CCDC93) and plays a broad role in intracellular trafficking and surface expression of various receptors and transporters[1][4][6]. All COMMD proteins—including COMMD8—form dimers and share structural features, supporting protein-protein interactions and complex assembly. While the family is involved in functions such as membrane trafficking, transcriptional regulation, and possibly metal homeostasis[1], most disease associations have been described for other family members (e.g., COMMD1 in copper metabolism disorders). There is currently no robust evidence to support COMMD8 as a direct therapeutic target, receptor, enzyme, or transporter, nor are there known drug interactions, mechanisms of action, or notable safety concerns described in the literature[2][6].
