Overview

COMM domain-containing protein 9 (COMMD9) is a member of the highly conserved COMMD protein family involved in the regulation of intracellular trafficking and transcription[2][3]. Structurally, it contains an α-helical N-terminal domain and a conserved C-terminal COMM domain, which enables the formation of homo- and heterodimers as part of larger protein complexes[2][3]. COMMD9 is an integral scaffold subunit of the Commander (CCC) complex, which regulates endosomal and trans-Golgi network trafficking, ion and lipid homeostasis, cell cycle, and immune responses[1][2][4]. In cancer biology, COMMD9 displays context-dependent functions: while some COMMD family members act as tumor suppressors, COMMD9 is overexpressed in certain lung cancers and promotes cell cycle progression via TFDP-1/E2F1 and p53 pathways, enhancing oncogenic potential[2]. COMMD9 is predicted to be involved in sodium ion transport and cholesterol homeostasis. Moreover, mutations in COMMD9 have been associated with Ritscher-Schinzel syndrome[4]. To date, there are no known drugs or biomarkers specifically targeting COMMD9, and its value as a therapeutic target remains under investigation rather than established.
