Complement 5a Receptor (C5aR)

Target
C5aR
Molecular classification
G protein-coupled receptor
01

Overview

Complement 5a Receptor (C5aR), also known as CD88 or C5AR1, is a G protein-coupled receptor that binds the complement component C5a. It is widely expressed on immune cells and mediates many of the pro-inflammatory effects of C5a, including chemotaxis, cytokine secretion, and phagocytosis. C5aR activation leads to downstream signaling through Gαi proteins, ERK1/2 phosphorylation, calcium mobilization, and Rho activation. Because excessive complement activation contributes to various diseases, C5aR is an attractive drug target for anti-inflammatory therapies, particularly for autoimmune disorders and transplant rejection. Antagonists, including small molecules and monoclonal antibodies, are being developed to block receptor function and reduce inflammation.
