Complement factor D (CFD), also known as adipsin, is a serine protease that plays a critical role in the activation of the alternative pathway of the complement system. It is the rate-limiting enzyme in this pathway, controlling its activity and output. CFD catalyzes the cleavage of complement factor B when it is complexed with C3b, leading to formation of C3 convertases, which are essential for amplifying complement activation. Alterations in CFD levels or function have been implicated in several disease contexts, including increased susceptibility to infections when deficient and contribution to inflammatory diseases when overactive. Therapeutic targeting strategies include monoclonal antibodies against CFD or small molecule inhibitors designed for precision modulation.
