Complement receptor type 1 (CR1), also known as C3b/C4b receptor or CD35, is a membrane glycoprotein that plays a crucial role in the regulation of the complement system. It acts as a negative regulator by binding activated complement fragments C3b and C4b, accelerating the decay of complement convertases and serving as a cofactor for Factor I-mediated inactivation of C3b and C4b. CR1 is expressed on various immune cells and erythrocytes, facilitating immune complex clearance and preventing tissue deposition. Decreased expression or mutations in CR1 are associated with several autoimmune and inflammatory diseases, including systemic lupus erythematosus and Alzheimer's disease. A soluble form of CR1 (sCR1) has been developed as a therapeutic agent to inhibit unwanted complement activation.
