Complex II is a key enzyme complex in mitochondria, serving as a crucial link between the tricarboxylic acid (TCA) cycle and oxidative phosphorylation (OXPHOS). It catalyzes the oxidation of succinate to fumarate in the TCA cycle, transferring electrons from succinate to FAD, then through iron-sulfur clusters, and finally to ubiquinone, reducing it to ubiquinol. Unlike other respiratory complexes, Complex II does not pump protons directly. It has additional roles in ROS generation, apoptosis, and cellular signaling. Mutations or deficiencies in complex II subunits can cause severe metabolic diseases, including cancers and neurodegenerative disorders.
