Overview

Connector enhancer of kinase suppressor of ras 3 (CNKSR3) is a scaffold/adaptor protein primarily involved in the regulation of epithelial sodium channel (ENaC)-mediated sodium transport and aldosterone-induced responses in various tissues, particularly the kidney. CNKSR3 acts as a molecular scaffold to coordinate the assembly of multiprotein complexes regulating sodium transport across epithelial membranes, contributing to the maintenance of sodium balance and blood pressure. It also negatively regulates the ERK1/2 signaling pathway and modulates peptidyl-serine phosphorylation. Mutations or altered expression of CNKSR3 have been associated with rare conditions such as immunodeficiency 40 and xanthinuria. Currently, no drugs are known to interact directly with CNKSR3, and it is not used as a biomarker or implicated in direct safety concerns in therapeutic settings
