Overview

Connexin 43 hemichannels are hexameric assemblies of Connexin 43 proteins in the plasma membrane that, when undocked from adjacent cells, serve as gated channels for small molecules and ions between the cytoplasm and extracellular environment[1][2][3][4][5]. While normally closed under physiological conditions, these hemichannels can open in response to stress, injury, inflammation, or mechanical stimuli, modulating processes such as ATP and prostaglandin release, cell survival, and local signaling in tissues including heart, brain, and bone[1][2][3][5]. Dysregulation of Cx43 hemichannels has been implicated in various diseases such as neurodegeneration, cardiovascular conditions, bone disorders, and cancer, making them regarded as promising therapeutic targets, especially for interventions requiring control of inflammation, tissue repair, or mechanosensitive cell signaling[1][4][5]. Selective pharmacological or antibody-based inhibition of Cx43 hemichannels is under investigation as a means to ameliorate pathological channel opening while preserving physiological gap junction intercellular communication[1][5].
