Connexin43 (Cx43), encoded by the GJA1 gene, is a member of the connexin family of proteins that form gap junction channels and hemichannels in vertebrate cells. Cx43 is one of the most widely expressed and studied connexins, playing critical roles in intercellular communication across various tissues, including heart, bone, skin, and nervous system. It mediates direct cytoplasmic exchange between neighboring cells via gap junctions and allows communication between the cell interior and extracellular environment via hemichannels. Dysregulation of Cx43 is implicated in various diseases, making it a drug target for cancer therapy, wound healing modulation, cardiac arrhythmia prevention/treatment, and eye injury/inflammation management.
