A contact sensitizer is a chemical substance, typically a low-molecular-weight hapten, that possesses the potential to induce an allergic immune response upon skin contact (Martin SF, 2012; PMID: 22612390). These molecules are generally not immunogenic in their native state but acquire immunogenicity by covalently binding to endogenous skin proteins to form hapten-protein complexes (Kimber I, et al., 2018; PMID: 29550844). This process initiates the sensitization phase of allergic contact dermatitis (ACD), where dendritic cells process the complexes and present them to T lymphocytes in regional lymph nodes. Upon subsequent exposure, a rapid inflammatory response is triggered, leading to the clinical symptoms of ACD, such as erythema and edema (StatPearls, 2023; NBK441867). In the pharmaceutical and cosmetic industries, identifying contact sensitizers is a mandatory component of safety assessment to prevent occupational and consumer health risks (OECD, 2010; Test No. 429). Common examples include nickel, formaldehyde, and various fragrance allergens, which are screened using both in vivo assays like the Local Lymph Node Assay (LLNA) and modern in vitro alternatives.
