Overview

Contactin-1 is a glycosylphosphatidylinositol (GPI)-anchored cell adhesion protein in the immunoglobulin superfamily, encoded by the gene CNTN1[4]. It plays a critical role in the development and maintenance of the nervous system, organizing axon connections, supporting formation and maintenance of myelin sheaths, and mediating neuron-glia communication[5][1][3]. Contactin-1 forms a key adhesion complex with neurofascin 155 and Caspr, necessary for establishing paranodal axoglial junctions at nodes of Ranvier, thus regulating saltatory nerve conduction in myelinated nerves[3][5]. Dysfunction or deficiency of contactin-1 leads to disrupted myelination and neurological disorders, highlighting its major role in both central and peripheral nervous system health[5][4][3].
