Contactin-3 (CNTN3) is a member of the contactin subfamily of neural immunoglobulin (Ig) domain-containing cell adhesion molecules. It is located on the cell surface by a glycosylphosphatidylinositol (GPI) anchor and features an extracellular domain with six N-terminal Ig domains and four fibronectin type III domains. CNTN3 is mainly expressed in the nervous system and is involved in mediating cell surface interactions important for neuronal network growth, axon guidance, and brain development. While less studied than other contactin family members, it has been genetically implicated in neurodevelopmental disorders such as autism spectrum disorders. CNTN3 interacts with neuronal cell adhesion molecules and receptor protein tyrosine phosphatases, potentially as part of higher-order signaling complexes, but its distinct functional consequences are not fully elucidated. Currently, it is not regarded as a direct therapeutic target, and there are no known drugs that specifically modulate CNTN3.
