Overview

Contactin-6 is a cell adhesion molecule of the immunoglobulin superfamily, also known as NB-3, encoded by the CNTN6 gene[1][2]. It is a GPI-anchored membrane protein containing six immunoglobulin-like and four fibronectin type III domains. Contactin-6 plays a major role in the development and maintenance of neuronal connectivity, especially in the central nervous system. It governs processes such as axon guidance, neurite outgrowth, synaptogenesis, and cell adhesion. Contactin-6 interacts with various protein partners, notably forming complexes in cis with latrophilin-1 (an adhesion GPCR), and with molecules such as CHL1, Ptpra, and Notch, participating in pathways critical for neuronal survival and apoptosis modulation[1][2]. Genetic studies have associated alterations in Contactin-6 with neurodevelopmental disorders, including autism spectrum disorder and coordination defects. While of significant interest in neuroscience and disease research, Contactin-6 is not an established therapeutic target and has no approved interacting drugs[1][2].
