Controlled release depot

A controlled release depot is a drug delivery platform rather than a biological molecule, receptor, or enzyme. It consists of a specialized formulation—typically an injectable liquid, gel, or solid implant—that is administered into a localized area, such as muscle or subcutaneous tissue, to provide sustained release of a therapeutic agent over an extended period ranging from weeks to months (StatPearls, 2023). This system is designed to maintain therapeutic drug concentrations in the bloodstream, thereby improving patient adherence and reducing the side effects associated with the peak plasma levels of immediate-release medications (PubMed, PMC4124997). Common technologies used in depots include biodegradable polymers like poly(lactic-co-glycolic acid) (PLGA) or the use of lipophilic prodrug esters in oil-based vehicles (NIH, 2022). While not a therapeutic target itself, it is a critical technology in the management of chronic diseases such as schizophrenia, opioid dependence, and hormone-sensitive cancers, where consistent dosing is vital for efficacy.
