Overview

COP9 signalosome complex subunit 8 (COPS8) is the smallest of eight protein subunits forming the highly conserved COP9 signalosome (CSN) complex, which is a key regulator in multiple cellular signaling pathways. The CSN complex is structurally and functionally related to the 19S regulatory particle of the 26S proteasome and plays a critical role in regulating ubiquitin ligase activity, particularly by deneddylating cullin proteins within SCF-type E3 ubiquitin ligase complexes. This regulation is essential for controlling protein turnover and modulating signaling cascades. Mutations or dysfunction in COPS8 have been linked to several genetic disorders and may also play roles in cancer pathogenesis through effects on proteostasis and cell cycle regulation[1][2][4][8].
