Overview

Copine-4 (CPNE4) is a member of the copine family, which are highly conserved, calcium-dependent membrane-binding proteins characterized by their tandem C2 domains and a von Willebrand factor type A (VWA) domain[2]. Copine-4 is predominantly expressed in the brain, especially in retinal ganglion cells and excitatory neurons, and functions as a calcium sensor involved in transient plasma membrane association, cytoskeletal interactions, and protein–protein binding[1][2][3]. It regulates neuronal morphology and differentiation, is linked with endolysosomal pathways and synaptic processes, and may participate in cellular responses related to neurodevelopment and neurodegeneration, such as Alzheimer’s disease[1][2]. Though not currently considered a direct therapeutic target or receptor, evidence implicates Copine-4 in several brain disorders and cancer[1][2].
