Overview

Copine-6 (CPNE6) is a brain-specific, calcium-dependent phospholipid-binding protein that belongs to the copine family and contains two N-terminal C2 domains and a von Willebrand factor A domain[4][7]. It is a mediator of synaptic plasticity, translating neuronal calcium transients into structural changes of dendritic spines by recruiting actin-modulating proteins such as Rac1 and Cofilin to postsynaptic membranes[1][2][3][5]. Copine-6 additionally interacts with key synaptic proteins, including VAMP2 and supports NMDA receptor-regulated signaling[5][6]. Loss or dysregulation of Copine-6 impairs long-term potentiation, learning, and memory, and has been associated with pathological brain conditions such as epilepsy[3][5]. Copine-6 is not presently considered a direct therapeutic target nor are there compounds targeting it in current clinical use.

Mechanism of action

For jasplakinolide, acts downstream by stabilizing actin to compensate for the lack of Copine-6-mediated actin regulation. No drugs directly targeting Copine-6 are described in the literature.
