Copine-8 is a highly conserved, soluble, calcium-dependent membrane-binding protein that participates in molecular events at the interface of the cell membrane and cytoplasm. Structurally, it contains two N-terminal C2 domains that bind calcium and phospholipids, and a C-terminal A domain similar to integrins for protein interactions. Its roles include mediating membrane trafficking, protein recruitment, lipid signaling pathways, and immune modulation. The gene is implicated in cancer progression as a prognostic and immunological biomarker in multiple malignancies, and in prion disease as a candidate gene affecting disease incubation time in mice. Its precise functions and clinical value are still being elucidated.
