Overview

Cordon-bleu WH2 repeat protein-like 1 (COBLL1) is an evolutionarily conserved actin-binding protein characterized by the presence of a WH2 (Wiskott–Aldrich syndrome protein homology 2) domain. It regulates actin cytoskeleton dynamics and is involved in morphogenic changes in cancer cells, promoting cell growth, migration, and morphogenesis—particularly in castration-resistant prostate cancer, where it supports androgen receptor phosphorylation and function[1]. In hematologic malignancies such as chronic lymphocytic leukemia, COBLL1 is a binding partner of the receptor ROR1 and serves as an independent prognostic biomarker, affecting cell signaling and migration. High expression of COBLL1 is associated with more aggressive disease course and alterations in cell motility and response to microenvironmental cues[1][2]. Despite these roles, no approved drugs directly target COBLL1 as of current scientific knowledge.
