The CoREST complex is a multi-protein transcriptional corepressor complex that plays a central role in the regulation of gene expression through chromatin modification. It is unique for containing both histone demethylase and deacetylase activities within the same assembly. Key components include RCOR1/2/3, LSD1/KDM1A, and HDAC1/2. It functions in transcriptional repression, particularly of neuron-specific genes, and is implicated in cancer and neurodevelopmental disorders.
Inhibition of histone demethylation (H3K4me/me2) and histone deacetylation (H3K9ac) to promote chromatin condensation and gene repression.
