The corneal epithelial glycocalyx is a specialized mucin-rich layer located at the interface between the corneal epithelium and the tear film. It primarily consists of membrane-associated mucins (such as MUC1, MUC4, and MUC16) and secreted mucins (like MUC5AC) that provide a hydrophilic environment, ensuring the tear film adheres to the hydrophobic ocular surface (Gipson, 2004). This layer serves critical roles in lubrication, hydration, and acting as a physical and chemical barrier against pathogens and environmental insults (Mantelli & Argüeso, 2008). In conditions like dry eye disease, the glycocalyx is often compromised, leading to tear film instability and epithelial damage (Koh, 2020). Therapeutic interventions often aim to restore this layer by stimulating mucin production or providing mucin-mimetic substances to improve ocular surface health (Urashima et al., 2004). By maintaining the integrity of this mucin-like layer, drugs can effectively reduce symptoms of irritation and prevent further damage to the corneal surface.
