Overview

Corneal nerves are the primary sensory nerves innervating the ocular surface, especially the cornea. They originate mainly from the ophthalmic branch of the trigeminal nerve (cranial nerve V), with their cell bodies located in the trigeminal ganglion[1][3][4]. The cornea is among the most densely innervated tissues in humans, containing thousands of free epithelial endings per square millimeter[3][4]. These nerves are responsible for sensing temperature, chemical and mechanical stimuli—including pain—and play a critical role in maintaining ocular surface homeostasis by mediating protective reflexes such as blinking and tear secretion[1][3]. Damage or dysfunction of these nerves can lead to significant clinical problems including dry eye disease, neurotrophic keratopathy, impaired wound healing, and neuropathic ocular pain syndromes[1][4]. Recent advances include drugs that promote regeneration of these nerves to address unmet therapeutic needs related to their injury or degeneration[1]. Note: "Ocular surface nerves" is not a canonical molecular target but rather refers collectively to peripheral sensory neurons innervating the ocular surface. It is not a single molecule/receptor but an anatomical/functional entity composed primarily of branches from cranial sensory neurons. Therefore, it does not fit standard drug target classifications such as receptor or enzyme; this makes "is_target" false and "is_incorrect" true for structured drug discovery purposes.
