Cortactin-binding protein 2 (CTTNBP2) is a neuron-specific scaffolding protein characterized by six ankyrin repeats and multiple proline-rich domains[3][1]. It is predominantly expressed in the brain where it is highly concentrated in dendritic spines, regulating their formation and maintenance by directly interacting with cortactin – a key F-actin branching factor – and components of the phosphatase 2A (PP2A) complex[1][2][3]. CTTNBP2 organizes the actin cytoskeleton and microtubules, with functional modules mediating its respective interactions. It plays a critical role in neuronal morphogenesis; knockdown in neurons impairs dendritic spine density and arborization[1][2]. Variations in the CTTNBP2 gene are associated with autism spectrum disorder, suggesting a significant role in neural development and synaptic signaling[2]. CTTNBP2 is not known as a direct therapeutic drug target, nor are there known small-molecule modulators or biomarker applications at present[1][2][3].
