Cortical neurons in the prefrontal cortex (PFC) are the primary cellular components of the brain region responsible for executive functions, including working memory, impulse control, and decision-making [Miller & Cohen, 2001]. This population consists mainly of excitatory glutamatergic pyramidal neurons and various classes of inhibitory GABAergic interneurons that work in concert to regulate cognitive and emotional processes [Arnsten, 2009]. Dysregulation of these neurons, often characterized by altered synaptic plasticity or an imbalance between excitation and inhibition, is a central feature of many neuropsychiatric disorders such as schizophrenia, ADHD, and major depression [Goldman-Rakic, 1995]. While "cortical neurons" is a cellular classification rather than a single molecular target, these cells express a high density of clinically relevant receptors, including NMDA, D1 dopamine, and 5-HT2A serotonin receptors [Stahl, 2021]. Pharmacological agents target these specific receptors to modulate the activity of PFC neurons, aiming to restore normal cognitive function or alleviate psychiatric symptoms [Logue & Gould, 2014]. Consequently, understanding the physiology of these neurons is vital for the development of next-generation neurotherapeutics.
Drugs interact with specific receptors (e.g., NMDA, Dopamine, Serotonin) expressed on these neurons to modulate excitatory and inhibitory neurotransmission, thereby altering the functional connectivity and firing patterns of the prefrontal circuitry [Stahl, 2021; Arnsten, 2009].
