Overview

Corticosteroid 11-beta-dehydrogenase isozyme 1 (11β-HSD1) is an NADPH-dependent enzyme that catalyzes the reversible conversion of inactive cortisone to active cortisol. It is highly expressed in metabolic tissues including the liver, adipose tissue, and central nervous system. By regenerating active cortisol within tissues, it amplifies local glucocorticoid action independently of circulating hormone levels. Overactivity or dysregulation has been implicated in obesity-related complications and cognitive symptoms linked with stress axis overactivation.
