The COVID-19 3C-like protease (3CLpro), also known as the main protease (Mpro) or nonstructural protein 5 (nsp5), is a cysteine protease essential for SARS-CoV-2 replication. It cleaves viral polyproteins pp1a and pp1ab at eleven conserved sites, releasing nonstructural proteins (nsps) required for forming the replication-transcription complex. Inhibition of 3CLpro blocks viral polyprotein processing, halting virus replication, making it a key target for antiviral drug development.
