Overview

Coxsackievirus A10 (CV-A10) RNA-dependent RNA polymerase (3Dpol) is the essential enzyme responsible for the replication and transcription of the viral RNA genome (PubMed: 31434734). As a member of the Enterovirus genus within the Picornaviridae family, CV-A10 is a major causative agent of hand, foot, and mouth disease (HFMD) and herpangina, which can lead to severe clinical manifestations in children (PubMed: 29151270). The 3Dpol enzyme catalyzes the synthesis of both negative-strand RNA templates and positive-strand genomic RNA, utilizing a protein-primed mechanism (UniProt: Q66578). Structurally, it adopts a conserved right-hand fold with fingers, palm, and thumb domains, where the palm domain contains the catalytic active site (PubMed: 31434734). Since RNA-dependent RNA polymerases are not found in human cells, 3Dpol is a primary target for the development of selective antiviral agents. Therapeutic approaches include the use of nucleoside analogs such as Ribavirin and Favipiravir, which function by causing premature chain termination or lethal mutagenesis during viral RNA synthesis (PubMed: 30209175).
