CRACD-like protein (CRACDL) is a protein encoded by the CRACDL gene (formerly C2orf55, KIAA1211L) on chromosome 2q11.2 in humans[1][2][3]. It is highly expressed in the cerebral cortex and in other tissues such as hippocampus, lung, pancreas, and kidney[1][2]. The protein, approximately 962 amino acids in length, is proline-rich and contains a highly conserved DUF4592 domain. CRACDL localizes mainly to the nucleus, microtubules, and centrosomes, with a predicted nuclear localization signal at residues 25–43[1][2]. Post-translational modifications include phosphorylation (Ser92, Ser490) and SUMOylation at five lysine residues[1][2]. CRACDL is predicted to interact with proteins associated with microtubule function (such as GSK3B, which is implicated in neurodegenerative and psychiatric disorders), but the precise biological functions remain incompletely characterized[1][2]. Clinically, CRACDL has been linked to several psychiatric conditions (depression, bipolar disorder, and schizophrenia) and various cancers (notably breast and ovarian cancers)[1][2][3]. The protein does not currently have known drugs targeting it, nor is it classified as a receptor, enzyme, transporter, or traditional therapeutic target. Its main classification is as a nuclear and cytoskeletal-associated protein containing a domain of unknown function (DUF4592)[1][2].
