In cancer, CRTC1 can act as a driver when fused to MAML2, forming a potent oncogenic fusion transcript implicated in mucoepidermoid carcinoma[2][3]. Loss or dysregulation of CRTC1 activity is implicated in neurodegenerative diseases like Alzheimer's disease, underscoring its essential role in neural gene expression and survival[4].
