Overview

CRK antisense RNA 1 (CRK-AS1) is a long noncoding RNA gene that functions as an antisense RNA, expressed from the opposite strand of the protein-coding CRK locus[1][7]. Antisense RNAs regulate gene expression through multiple mechanisms, including transcriptional interference, epigenetic modifications, and post-transcriptional RNA duplex formation[2][8]. These regulatory RNAs typically act in cis (at their site of synthesis) or trans (across different chromosomal loci), influencing their sense gene or nearby genomic regions[2]. While many antisense long noncoding RNAs have been implicated in various cancers and cellular processes such as proliferation and apoptosis[8], there is currently no direct evidence that CRK-AS1 itself serves as a therapeutic target or biomarker. Its biological significance remains an active area of research, especially in the context of epigenetic regulation and gene expression[1][2][8]. In summary, CRK-AS1 is a noncoding regulatory RNA, not a canonical receptor, enzyme, or therapeutic target, and is classified within the growing family of antisense long noncoding RNAs.

Biological functions

Regulation of gene expression (acts in cis or trans to modulate nearby genes, mainly the sense CRK locus)Epigenetic modification (sometimes recruits chromatin-modifying enzymes for gene regulation, as described for other antisense RNAs)Potential involvement in cell proliferation and apoptosis (based on lncRNA and antisense RNA studies; specific CRK-AS1 roles in these processes remain unclear)
03

Disease associations

Cancer (many antisense lncRNAs exert regulatory roles in cancer, though specific evidence for CRK-AS1 is limited to suggesting involvement due to its general class; no specific clinical associations yet for CRK-AS1)Other: No validated roles in other diseases to date
